Stora Mellösa kyrka is a medieval stone church in Stora Mellösa, Örebro kommun, serving today as the parish church of Kvismare församling within the Diocese of Strängnäs. Its long history stretches back to the twelfth century.

The church was originally built as a stone structure in the later 1100s, making it one of the older churches in the district. It was substantially enlarged in the fifteenth century, when the building was extended eastward and given two vaulted aisles. A sacristy and porch were added around the same period or slightly later, in the fifteenth or sixteenth century.

In the mid-1600s, the Göksholm family funerary chapel was built onto the church, reflecting the involvement of local landed gentry in its fabric. Disaster struck on 26 May 1743, when lightning hit the building, destroying the spire, roof and parts of the interior fittings—a reminder of the everyday hazards faced by rural churches before modern lightning protection.

The most dramatic transformation came between 1830 and 1836, when the entire church was rebuilt. The medieval vaults were demolished and replaced, and a semicircular chancel was added, giving the building much of its present external appearance.

Among the church's furnishings, the Romanesque font dates from the twelfth century, making it older than most of the current building fabric and a survivor of the church's earliest phase. The altarpiece was created in 1712–1713 by Olof Malmström of Stora Malm in Södermanland, while the pulpit dates from the 1830s rebuilding. Silver communion vessels and other fittings span several centuries, including a wine jug from 1651 and altar candlesticks added as late as 1961.

The church has had a succession of organs since one was first donated in 1615; the current gallery organ, built in 1919 by E. A. Setterquist & Son of Örebro and later modified, retains a façade from an earlier eighteenth-century instrument by Pehr Zacharias Strand. A separate chancel organ was added in 1986.

In the churchyard stands the runestone known as Nä 12, and another, Nä 13, is built into the wall of the porch—tangible links to the site's pre-Christian and early medieval past.
